ScienceDaily (Feb. 24, 2012) ? Researchers at the RIKEN-MIT Center for Neural Circuit Genetics have discovered an answer to the long-standing mystery of how brain cells can both remember new memories while also maintaining older ones. They found that specific neurons in a brain region called the dentate gyrus serve distinct roles in memory formation depending on whether the neural stem cells that produced them were of old versus young age.
The study will appear in the March 30 issue of Cell and links the cellular basis of memory formation to the birth of new neurons -- a finding that could unlock a new class of drug targets to treat memory disorders.
The findings also suggest that an imbalance between young and old neurons in the brain could disrupt normal memory formation during post-traumatic stress disorder (PTSD) and aging. "In animals, traumatic experiences and aging often lead to decline of the birth of new neurons in the dentate gyrus. In humans, recent studies found dentate gyrus dysfunction and related memory impairments during normal aging," said the study's senior author Susumu Tonegawa, 1987 Nobel Laureate and Director of the RIKEN-MIT Center.
Other authors include Toshiaki Nakashiba and researchers from the RIKEN-MIT Center and Picower Institute at MIT; the laboratory of Michael S. Fanselow at the University of California at Los Angeles; and the laboratory of Chris J. McBain at the National Institute of Child Health and Human Development.
In the study, the authors tested mice in two types of memory processes. Pattern separation is the process by which the brain distinguishes differences between similar events, like remembering two Madeleine cookies with different tastes. In contrast, pattern completion is used to recall detailed content of memories based on limited clues, like recalling who one was with when remembering the taste of the Madeleine cookies.
Pattern separation forms distinct new memories based on differences between experiences; pattern completion retrieves memories by detecting similarities. Individuals with brain injury or trauma may be unable to recall people they see every day. Others with PTSD are unable to forget terrible events. "Impaired pattern separation due to the loss of young neurons may shift the balance in favor of pattern completion, which may underlie recurrent traumatic memory recall observed in PTSD patients," Tonegawa said.
Neuroscientists have long thought these two opposing and potentially competing processes occur in different neural circuits. The dentate gyrus, a structure with remarkable plasticity within the nervous system and its role in conditions from depression to epilepsy to traumatic brain injury -- was thought to be engaged in pattern separation and the CA3 region in pattern completion. Instead, the MIT researchers found that dentate gyrus neurons may perform pattern separation or completion depending on the age of their cells.
The MIT researchers assessed pattern separation in mice who learned to distinguish between two similar but distinct chambers: one safe and the other associated with an unpleasant foot shock. To test their pattern completion abilities, the mice were given limited cues to escape a maze they had previously learned to negotiate. Normal mice were compared with mice lacking either young neurons or old neurons. The mice exhibited defects in pattern completion or separation depending on which set of neurons was removed.
"By studying mice genetically modified to block neuronal communication from old neurons -- or by wiping out their adult-born young neurons -- we found that old neurons were dispensable for pattern separation, whereas young neurons were required for it," co-author Toshiaki Nakashiba said. "Our data also demonstrated that mice devoid of old neurons were defective in pattern completion, suggesting that the balance between pattern separation and completion may be altered as a result of loss of old neurons."
The work was supported by the RIKEN-MIT Center for Neural Circuit Genetics, Howard Hughes Medical Institute, Otsuka Maryland Research Institute, Picower Foundation and the National Institutes of Health.

?Sure, the truth is that there are no drugs FDA approved for this rare cancer, and there is no drug known to be particularly efficacious,? Fisher said. ?But to stand by, do nothing, offer no treatment, and simply watch this child die from cancer is against the standard of care for children with cancer in the United States.?
Fisher wrote to Nevada?s congressional delegation, state social services officials and the Reno Gazette-Journal, calling the decision not to approve treatment an example of a ?death panel.?
Amerigroup?s medical director subsequently approved Brandon?s course of treatment.
Eric Lloyd, the chief operating officer for Amerigroup Nevada, said the company understands the urgency of the boy?s case. He said payment approval for the first round of chemotherapy was denied because it was part of a clinical study and as such was ?experimental.?
In the current situation, he said, Fisher was able to show that the drugs had been used on patients with tumors like Brandon?s and the latest treatment would not be part of a clinical trial. Therefore, the therapy was approved for payment, he said.
?We?re doing everything we can to care for Brandon in every way we can,? Lloyd said.
Chuck Duarte, administrator for the Nevada Division of Health Care Financing and Policy, said Brandon?s case underscores a need to change Medicaid law.
?There needs to be a provision that when no approved therapy exists, doctors may show good cause to use what they think may help,? he said.
On Monday, Mojica-Guevara and his wife, Ana Rosa, tickled Brandon after giving the boy the liquid drugs through a tube in his nose. In a week or so, mother and son will be back at Stanford for radiation treatments.
?We?re praying for a miracle,? Mojica-Guevara said. ?At least now he has a chance.?

LLC vs. S Corp: Which Is Right for Your Startup?
Choosing the right business structure can be a daunting task for the small business owner or entrepreneur. You may have heard that the traditional C Corporation is overkill for most small businesses, and results in higher overall tax payments through something known as double taxation. But if the C Corporation isn?t right, then what is?
In my last post, I discussed how the LLC (limited liability company) and S Corporation are popular structures for small businesses since they avoid this double taxation burden. With these business structures, the company is taxed like a sole proprietor or partnership, meaning the company itself doesn?t file its own taxes; all company profits are ?passed through? and reported on the personal income tax return of the shareholders or, in the case of an LLC, the members.
Most importantly, both the LLC and S Corp will separate your personal assets from any liabilities of the company (whether from an unhappy customer, unpaid supplier, or anyone else who might pursue legal action).
The similarities between these two business entities are significant, but the differences can be even more striking. While circumstances vary for each individual and his or her business, here are some general guidelines to help you understand the differences and their impact on your business.
1. Business Formality
With its roots as a C Corporation, the S Corporation involves structure, formalities and compliance obligations, which can be too burdensome for the solo entrepreneur, in other words, a ?payroll of one.? If you incorporate as an S Corporation, you need to set up a board of directors, file annual reports and other business filings, hold shareholder?s meetings, keep records of your meeting minutes, and generally operate at a higher level of regulatory compliance than your business might need or want to deal with. With the LLC, this isn?t the case. LLCs just use an informal operating agreement.
What to know: If you want less red tape and formality, the LLC can provide greater simplicity.
2. Who Can Be a Shareholder?
The S Corporation has more restrictions in terms of who can be a shareholder. For example, an S Corp cannot have more than 100 shareholders. Of course, this limitation is probably not of much consequence to many small businesses. In addition, all individual shareholders of an S Corp must be either U.S. citizens or permanent residents.
What to know: If you have foreign owners (or would like an LLC to be a shareholder), you cannot form an S Corporation and should opt for the LLC.
3. Income Allocation
?In an LLC, income and loss can be allocated disproportionately among the owners. By contrast, in the S Corp, income and loss are assigned to each shareholder strictly based on their pro-rata shares of ownership.
This is an important distinction to understand. For example, let?s say Sally and Heidi open a micro-brewery, each owning 50%. As the year progresses, Sally needs to focus her time elsewhere, while Heidi does all the work. Their business becomes more profitable than they ever imagined, and they want to divide up the profits. Because Heidi has put in the bulk of the work, the two decide she should keep 75% of the profits, and Sally should get 25%.
With an LLC, this type of agreement is fine. The two owners simply agree to the arrangement and they will be taxed accordingly to their ?operating agreement.?
But this type of flexible arrangement won?t work with an S Corporation. Because Sally and Heidi are each 50% owners, each will be allocated 50% of the corporation?s income, at least when it comes to computing income tax.
What to know: If you need flexibility when it comes to divvying up profits among owners, the LLC is the preferred structure.
4. Pass-Through Losses
With LLCs and S Corporations, members and shareholders are able to pass company losses to their personal income reporting. In some circumstances, the LLC lets you pass more loss than in an S Corporation, most notably when it comes to real estate. In an LLC used for real estate investments, members are allowed to add the amount of the mortgage to their basis for the purpose of computing a loss. Clearly, that can add up to a significant difference in your tax statement.?
What to know: If you?re setting up a business structure for real estate investments, the LLC lets you write off more losses on your personal tax reporting.
5. Class of Stock
In an S Corporation, all shareholders own only one class of stock. An S Corporation can have voting and non-voting shares, but cannot have distinctions like common stock and preferred stock. In an LLC, however, these priorities and preferences are allowed, and you can have different membership classes.
What to know: You cannot offer common and preferred stock classes in an S Corporation. Thus, if you?d like flexibility in ownership classification, go with the LLC.
6. Reinvesting Profits
There?s another twist regarding the LLC, S Corp and your taxes. As pass-through entities, individual owners of an S Corporation or LLC are liable for any taxes owed on profits ? whether that money is retained in the company or put in their wallets.
For example, if you own 50% of an S Corporation or LLC and that company makes $80,000 in profit, you need to report $40,000 in income on your personal tax return. And it doesn?t matter whether that $40,000 actually ended up in your pocket. This is known as ?phantom income,? and can obviously cause a problem for some shareholders.
What to know: If you plan on retaining money in the company (and would prefer not to have shareholders be personally taxed on this money), you should consider the C Corporation over both the LLC and S Corp. Of course, your specific situation may vary, so it?s always best to consult your accountant.
7. VC Funding
If your company is considering raising venture capital down the road, VC firms will most likely choose the C Corporation as the type of legal entity for their investments. This doesn?t necessarily mean your business needs to start as a C Corp, but be advised: If you are considering raising venture capital and start out with an LLC or an S Corp, you will need to convert the business to a C Corp (if your state allows conversions) at some point. This conversion will require additional filings and fees within your state. If you choose this route, you may want to consider the S Corp as your option, since converting an S Corp to a C Corp can be done in a day with a single tax form (you?re basically unchecking the box for S Corp tax election on an IRS form).
Choosing the right business structure is a multi-faceted decision, and will ultimately depend on all the unique aspects of your particular business needs, vision and circumstances. And remember that tax treatment varies between states.
Consulting with an accountant or tax advisor can go a long way in helping you determine which business structure offers the biggest advantage for your situation. Take note that the deadline to elect S Corp treatment is March 15 for existing companies (or 75 days from the day your company is formed). Get your legal structure squared away early on, and your company will be set for years to come.

Capitol Hill weighs GOP payroll tax gambit (AP)
WASHINGTON ? House lawmakers are returning to Capitol Hill to weigh a proposal by GOP leaders to extend the 2-percentage-point cut in the payroll tax through the end of the year and add the approximately $100 billion cost to the nation's $15 trillion-plus debt instead of scrounging around the budget for ways to pay for it.
In the Democratic-majority Senate, a top leader said the proposal was "a major step forward" even as other Democrats worried it could jeopardize efforts to renew unemployment benefits for millions of the long-term jobless and efforts to forestall a scheduled cut in fees to doctors who treat Medicare patients. Those proposals would remain in the hands of a House-Senate negotiating panel that's looking for spending cuts to defray the deficit impact.
The idea is sure to be a topic of conversation Tuesday at the weekly closed-door conferences of Senate Democrats and Republicans.
House Speaker John Boehner, R-Ohio, Majority Leader Eric Cantor, R-Va., and GOP Whip Kevin McCarthy of California said the House could vote on the payroll tax measure this week but the renewal of jobless benefits and the Medicare "docs' fix" still would have to be paid for with spending cuts elsewhere.
The GOP statement came after intense talks over the weekend failed to produce an agreement. Republicans were pressing for pay cuts for federal workers and requiring them to contribute more to their pensions. They recoiled at a Democratic proposal to raise Transportation Security Administration per-ticket airline security fees.
"Democrats' refusal to agree to any spending cuts in the conference committee has made it necessary for us to prepare this fallback option to protect small business job creators and ensure taxes don't go up on middle-class workers," the GOP leadership statement said.
Without action by Congress by the end of the month, payroll taxes will rise for 160 million Americans. The 2-percentage-point tax cut delivers about $20 a week to a worker making $50,000 a year and a tax cut totaling $2,000 this year for someone making a $100,000 salary.
Democrats were encouraged and said the development could break an impasse over the payroll tax proposal and the other expiring provisions.
"We've been making the point that when (it comes to) tax cuts for folks at the very top, the House Republicans went to great lengths to change their rules to say you don't have to pay for those," said Rep. Chris Van Hollen, D-Md. "And yet they've been saying that when it comes to a short-term, 10-month payroll tax cut for middle-income people, all of a sudden you have to pay for it."
But Democrats warned that decoupling the payroll tax from the larger legislation could jeopardize efforts to renew the jobless benefits and the fix to the Medicare payment formula.
"It's completely irresponsible to leave behind nearly 5 million unemployed Americans whose benefits will expire and 47 million seniors and disabled Americans whose access to health care would be jeopardized," said Rep. Sander Levin, D-Mich., a member of the 20-lawmaker House-Senate negotiating panel.
"There is no reason all three of these priorities cannot proceed at the same time," House Minority Leader Nancy Pelosi, D-Calif., said.
The GOP move reflects a desire by party leaders to avoid a political hit if the payroll tax expires at the end of the month. And it would avoid burdening businesses with uncertainties regarding their payroll systems. On the other hand, jobless benefits lapsed for several weeks in 2010, and delays in adopting the Medicare fix can be dealt with by delaying the processing of Medicare claims.
"It is prudent for our leadership to take whatever action is necessary to ensure American workers are not hit with a tax increase on March 1," said Rep. Dave Camp, R-Mich., the lead GOP negotiator.
The White House did not embrace the House leadership idea.
"We are willing to work with them to offset it in a responsible way," White House press secretary Jay Carney said. "And we expect Congress to get its work done and to extend it ? the payroll tax cut, unemployment insurance and the doc fix." But Carney added, "Let's just see how this process plays out."
The move by the GOP leadership still would leave it to negotiators to come up with $30 billion or $40 billion in deficit savings to extend jobless benefits averaging about $300 a week to people who have been out of work for more than six months. Republicans have pressed to cut the number of weeks from the maximum 99 permitted under current policies and economic conditions down to as few as 59 weeks. They also are pressing to require people receiving unemployment to enroll in GED classes and allow states to condition benefits on the passage of drug tests.
